First leaf has spots

I took a cutting from a friend's plant a while ago and had it in a cup of water for a few weeks until it grew roots. I put it in some well draining soil and after a couple weeks it busted out this new leaf two days ago. Should I be concerned about the spots on the leaf? Thank you!
